1A revolution in olive oil analytics
Nuclear Magnetic Resonance (NMR) spectroscopy is a highly sophisticated, non-destructive analytical method. It relies on the absorption of radiofrequency radiation by active atomic nuclei (primarily ¹H and ¹³C) when placed inside an extremely strong external magnetic field.
For extra virgin olive oil, NMR represents a complete paradigm shift compared to old-school wet chemistry methods: * Non-destructive: The sample remains intact and can be recovered after the test. * Ultra-fast: A complete, comprehensive analysis is finished in 5 to 15 minutes. * Highly informative: A single ¹H spectrum provides a complete chemical "fingerprint," revealing fatty acid profile, exact purity, and—using advanced statistical models—geographic provenance.
The official application for olive oil is regulated by the International Olive Council under the COI/T.20/Doc. No 30 protocol.
2How ¹H-NMR works: Mapping the molecules
To run the analysis, the olive oil is dissolved in deuterated chloroform (CDCl₃). When placed in the spectrometer, different chemical groups generate distinct peaks at specific positions (chemical shifts, measured in ppm): * Protons on double bonds (vinilic, δ 5.20–5.45 ppm): Directly proportional to total unsaturation. It distinguishes the ratios of monounsaturated (oleic) and polyunsaturated (linoleic, linolenic) fatty acids. * Bis-allylic protons (δ 2.75 ppm): The ultimate diagnostic peak for linoleic acid (omega-6). Its integration allows direct, high-precision quantification. * Allylic protons of linolenic acid (δ 2.05 ppm): Quantifies linolenic acid (omega-3), crucial for detecting blending.
3Geographic Fingerprinting and Fraud Detection
1. Spotting cheap seed oils Adulteration with seed oils (like high-oleic sunflower, corn, or hazelnut oil) is easily caught. For instance: * **Sunflower oil** spikes the bis-allylic peak (linoleic). * **Corn oil** displays a distinct signature of the plant sterol **beta-sitosterol** in the aliphatic region. * The detection threshold for seed oil mixing is below **1%** when combined with multivariate chemometric models.
2. Certifying authentic DOP and origin Because the micro-composition of triglycerides is shaped by cultivar, soil chemistry, and local weather patterns, the NMR spectrum of an olive oil serves as a geographic passport. By comparing a sample's spectrum against official databases, laboratories can verify if an oil labeled as "100% Tuscan DOP" actually matches the authentic regional signature.
